2020年年初，新冠肺炎疫情暴发，1月23日，武汉关闭离汉通道。900万武汉人与来自全国各地医疗系统及其他行业的一线人员，共同打响武汉抗击新冠肺炎疫情阻击战 。 《武汉日夜》剧照2020年4月，曹金玲接受导演工作后，反复观看已有的1000多个小时视频素材，终于定下《武汉日夜》主题是“陪伴”，根据这一主题进行梳理，让人物一组一组出现，因为病患与疾病的抗争也都是在家人、医生的鼓励下进行的。且这几条线也相对完整，具有一定的代表性，便于人物的立体呈现。 李玮表示，选取哪些人物在大银幕上呈现，这一过程持续了10个月，每版都不一样，一直到第四版才出来大致的框架 。与一般纪录片是从策划开始，然后拍摄，再后期制作不同，该片的素材直接来自当时冲在抗击疫情第一线的武汉媒体已有的新闻素材。因疫情原因，2020年疫情高峰期摄制组进不去现场。从1月份开始，电影频道做了大量与抗击疫情相关的节目，包括《最美的平凡》、《两地书》、《战疫故事》、《阳台里的武汉》等，在这个过程中，电影频道累积了不少视频素材 。
